Great paper demonstrating the extension of lifespan in wild-type mice by in vivo reprogramming! Although this is a great first step, I believe we are all waiting for a much stronger effect once we overcome the technical barriers. Let’s keep working together to get there!
Our new article reveals that gene therapy-mediated partial reprogramming extends lifespan and reverses age-related changes in aged mice, showcasing lifespan extension in wild-type animals through in vivo reprogramming.

Agree 100%. Very disappointing to see most "longevity" companies pivoting to obesity and disease, not even age-associated diseases anymore. We all know you need an indication, business model, investors, FDA, but this is clearly not the type of trials we all signed up to see!
Obesity is the hot topic in biotech/pharma right now. Alas, I fear this may be a bad thing for the aging field, something that sidetracks the field.
I'm prob one of the few not to fully celebrate BioAge's recent huge series D round. I'm good friends with the company & happy for

In my opinion, most clinical trials for the treatment of age-associated diseases by targeting fundamental mechanisms of aging will unfortunately fail! Although aging is the major risk factor for these diseases, it will be very difficult to treat them rather than prevent them!

When founders and investors think about the market size of drugs that target the biology of aging, we all need to undergo a complete change of mindset!
We found that a drug approved and indicated for aging would conservatively have a peak global market size ~$150-$200 billion annually.
A company that owned only this drug would be more valuable than the top 2 big pharma companies (J&J and Pfizer) in terms of revenue…combined.

We (as a field) have made very small progress during the last decade. In order to discover more and better drugs, we need to test more drugs!
A good way to assess if we are making progress in the field is to look at the best lifespan extension achieved so far in the ITP. So far, the best result for a single drug (excluding combinations) is for Rapamycin, in female mice, at 42ppm (+28%).

While there is association in mice between drugs than extend lifespan and loss of body weight (BW), there are examples of extension without BW loss, and BW loss without extension. What if loss of BW is a requirement for extension, but not necessarily the MOA?

I think that we are entering into a discussion about semantics. I agree that medicine extends lifespan of patients. In addition, if a drug can extend the lifespan of patient beyond the lifespan of healthy controls, this drug could potentially extend human lifespan.
