We haven't tested whether DNMT3L can prevent their loss, as this would require over-expression during or immediately after derivation of a new hTSC line. What we do see is that DNMT3L expression cannot recover imprinted methylation in hTSCs once lost.

Compared to early human trophoblast, hTSCs lack DNMT3Lexpression, which is necessary for PMDs in mouse trophoblast. When we ectopically express DNMT3L in hTSCs, we find placental PMDs are restored, supporting a conserved role for DNMT3L in de novo methylation in trophoblast.

Using single-cell RNA-seq + PBAT, we characterised the molecular profiles of NSN and SN GV oocytes, which show highly distinct chromatin structures (example below). A great collaboration!
