We observed that PD-1-induced inhibition of the actin was local, and that it didn't prevent immune synapse formation of the same cell on another spot.
Thus, we wondered if PD-1 signaling, depending on two previously described signaling motifs, was involved in the process.

To test the role of the motifs in actin defects, we mutated the 2 tyrosines and analyzed synapse formation in T cells expressing the WT or mutant PD-1.
This mutant was able to induce the same defects, showing the first reported PD-1 signaling motifs-independent inhibition.
