Beautiful work! Figure 7 is insightful. TME changes in response to tumor regression can be bystander events. To show CD8+ T-cell depletion partially hinders anti-tumor effect of KrasG12V targeting points to a function contribution. I am curious about the increase in M1-like cells

Using genetic screens, we nominate DAPK3 as a functional mediator of CRC differentiation following HDAC1/2 inhibition. These findings highlight the importance of chemically targetable histone modifications in controlling cellular states with therapeutic potential.

Genome-wide profiling assays revealed that H3K27ac and H3K9ac associated with open chromatin and upregulation of differentiation genes. Inhibiting EP300, a principle writer of H3K27ac, reversed CRC differentiation mediated by HDAC1/2 inhibition.

After confirming this finding across different CRC models, we profiled histone post-translational modifications, identifying 3 marks with the greatest abundance following HDAC1/2 inhibition.

Could not agree more! We need to support the entire spectrum of science, fundamental to translational. Funding basic science is critical as our predictive power for translation is limited.
