In this issue, we review the SHOC2-MRAS-PP1C (SMP) complex assembly, the substitution of MRAS by the canonical RAS proteins, and explore potential therapeutic approaches for targeting the SMP complex in RAS/RAF-driven cancers and RASopathies.
