@ProfluentBio This is incredibly important work. I've been hoping that a broader PAM "menu" would soon be available, since it can make a massive difference for the utility of base editors. It looks like you're moving the field in that direction, which is wonderful to see!

The Wilson lab has two new PhD candidates: 🍾 Congrats 🍾 to Brigette Manohar and Christy George on passing their @berkeleyMCB qualifying exams! They're both working to improve CRISPR delivery, potentially enabling new therapies for the central nervous system.

@rammydev @thisismadani @ProfluentBio The field typically uses E. coli to "print" proteins, transforming them from genetic code to a physical enzyme (which we can purify). In the work reported here, they put the coding DNA into human cells and let them do the same thing. So they are reporting a real-world test!

@budcolincoln @davidrliu @Nature @DDunkelmann If the entire "dictionary" of life's biochemistry had been only ~20 words, this work expands the vocabulary substantially. More words = proteins w/ novel/diverse functions. The next step will be to find ways to put these words into widespread use, which has been tricky so far.

@Ella_Maru @davidrliu @NatureBiotech A great thing about prime editing is that it doesn't rely on double-stranded breaks in DNA, which can result in chromosomal rearrangements which - although not necessarily dangerous - would be nice to avoid. And it relies on 3 steps: more of a "filter" preventing off-target edits

@Nicole_Paulk The terminology hills I'm ready to die on? CRISPR is not gene therapy - it's genetic therapy. Cutting w/ two gRNAs is not "dual guide" because that phrase = you used crRNA+tracrRNA (dgRNA). And it's technically CRISR-Cas9:

This is not a prank, but it's so foolishly pedantic that today is the perfect day to address the dark secret that many CRISPR researchers refuse to confront: Cas9 is *not* CRISPR. It is actually CRISR, because its locus is undeniably non-palindromic. Buckle up! #CRISPR #CRISR 1/n
