@WeiDu13 @erblabs @mpi_marburg And as a rough comparison/estimation, we routely cultivate E coli with only 10 mM glucose, which is for all the biomass/energy requirements, 0.5mM Thr should be enough to relieve auxotrophy due to the thrB knock out.

@WeiDu13 @erblabs @mpi_marburg For the Q2, nope, 0.5mM Thr should be enough. The strain also couldn't grow even when complemented with thrB from a plasmid (Fig. S9, black & blue lines).

@WeiDu13 @erblabs @mpi_marburg Thank you! For the Q1, mutations happened naturally/randomly, but the knock-out strain grew under selective conditions. We interpreted this to be a lower selection pressure when with PN. Fig4 indicated that 13nM could partially relieve selection.
