@WallaceUcsf @aaandmoore @Valm_Lab @cohenlaboratory @ericwait It would be totally cool to directly look at tether dynamics in the multispectral system and see if we can give more information on "who" is stable or not (even with an imperfect tether label)--but we've never gotten around to doing it in @JLS_Lab. šŸ˜•

@aaandmoore @WallaceUcsf End result: Colocalization in DL-imaging isn't great to define contact sites. Unfortunately, most of the tools for specifically labeling them aren't ideal either...it's a tough problem. Gold standard is still biochemistry and EM, so no easy dynamics.šŸ˜Ÿ

@AidanFenix @WallaceUcsf I think that only one part-thatā€™s the way we filter the T cell repertoire. My understanding is that the major way we detect and amplify an alarm signal is based on PRR stimulation.

Anybody else feeling some spaghetti-oh's? I'm sticking with the old school GI-SIM this week. Dense tubular(?) ER in a (very weird) COS7 cell, looks like sheet by DL imaging. Forgive the SIM artifacts, we were new to SIM in 2015 ;-) @AbellJonny
