Loss of PHRF1 results in significant up regulation of DDR and cell cycle checkpoint regulation pathways and down regulation of splicing and transcription-associated pathways. HeLa and HCT116 cells lacking PHRF1 are also highly sensitive to DNA damage. 3/

Specifically, we identify a single residue in the PHD finger that regulates histone binding, P221. We also show that PHRF1 associates with splicing factors and DNA damage repair proteins in cells. 2/
