We extensively tested IRescue by simulating 3' and 5' single cell/nuclei data, demonstrating its precision. We validated IRescue on four case studies of #TEs expression dynamics in humans: cancer, brain, skin aging and immune response to SARS-CoV-2 infection.

2/3 IRescue features a unique algorithm that handles UMI deduplication while accounting for sequencing errors, and optimally redistribute the counts from multi-mapping reads through an Expectation-Maximization approach.
