Last year we published the rationales to combine G12C inhibitors with anti-EGFR agents in colon cancer and the use of TDXd in HER2-mutant lung cancer. Very satisfying to see these results translated into clinical successes.

In conclusion, we show that KRAS G12C amplification is a frequent mechanism of resistance to G12C-EGFR blockade. Acquisition of an oncogene-induced senescence in these cells can be exploited as a therapeutic opportunity using senolytic agents.
