We conducted a pooled analysis of >200pts treated on 5 neoadj ADT+ARPI trials led by Mary-Ellen Taplin. Overall 5yr MFS (by conv scans) was 83%. RCB was strongly prognostic for MFS and we explored RCB categories to see association with MFS

These data have implications for treatment sequencing in mCRPC, particularly in DDR-altered patients for whom a PARPi may be available. We also await data from combo trials, incl our own #LuCarbo trial, evaluating DNA-damaging agents with LuPSMA.

@Dav_Lorente Thanks David! Absolutely, this work shows AI tools need external and ideally prospective validation before routine use - and that clinical parameters should also be included in these efforts

Though clinical/ICECAP risk should not be used to guide ADT duration in routine practice, it deserves evaluation in ongoing trials looking at ADT duration/intensification, and future digital path/AI studies should include comparison by these risk factors. @DanaFarber_GU @PCFnews
