And, thanks to reviewer urging, since our preprint, we added some functional studies, including for a gain-of-cys mutation in CAND1 that blocks interaction with Cul1 and mutations in HMGB1 that decrease cytokine activity

Overall, we remain very excited about the utility of covalent compounds as chemical probes and drugs, and we hope that by better understanding the mechanisms by which these compounds cause stress, we can help to ensure on-target activity and inform the functions of covalents. 5/5
